Abstract
Plants are a rich source of bioactive compounds with significant antioxidant potential, playing a vital role in preventing oxidative stress and decreasing the risk of chronic diseases like cardiovascular diseases, neurodegenerative disorders, and cancer. Evaluating the antioxidant properties of plants requires a diverse array of biochemical and molecular methodologies, including phytochemical screening, extraction techniques, and antioxidant activity assays. Key biochemical assays such as DPPH, ABTS, FRAP, and ORAC provide insights into the antioxidant capacity of plant extracts, while chromatographic and spectroscopic techniques, including HPLC, GC-MS, and FTIR, enable the identification and quantification of specific antioxidant compounds. Advanced analytical techniques, such as electrochemical, spectroscopic, fluorescence-based, and enzyme-based methods, further enhance the identification, quantification, and characterisation of antioxidants. Beyond antioxidant assessment, various experimental approaches have been explored to investigate the broader bioactive and therapeutic potential of plant-derived compounds. Cytotoxicity assays and enzyme inhibition studies help elucidate their pharmacological effects, including anti-inflammatory, neuroprotective, antimicrobial, and anticancer activities. Furthermore, genomic approaches such as transcriptomic and metabolomic analyses have emerged as powerful tools for understanding the genetic regulation of antioxidant biosynthesis in plants. Recent advances in biochemical assays and genomic technologies have significantly improved our ability to assess and enhance the antioxidant potential of plant compounds, with far-reaching implications for medicine, nutrition, and agriculture. This review comprehensively explores current methodologies for evaluating plant-based antioxidants, discusses their advantages and limitations, and highlights the integration of biochemical and genomic approaches in antioxidant research. A deeper understanding of these methodologies can drive innovation in functional food development, pharmaceutical formulations, and sustainable agriculture practices.
