Over the past 30 years, Streptozotocin (STZ) has been widely used to induce diabetes in animals and evaluate hypoglycemic drugs. However, the physicochemical properties and associated toxicity of STZ pose a challenge to researchers. Maintaining uniformity, consistency, reproducibility, and minimal animal mortality in STZ-induced diabetes is also challenging. Proper use of STZ is essential to prevent increased mortality and suffering in animals, and several factors must be considered, such as STZ production method, stability, appropriate dosage, route of administration, diet, animal species, age, body weight, sex, and target blood glucose level. The protocol for STZ-induced diabetes in experimental animals must be carefully developed, and this review provides an overview of diabetogenic mechanisms, specific toxicity, and factors leading to changes in STZ's diabetogenic effects.
