Abstract: In the past decade, the antimicrobial properties of plant extracts have gotten more attention. The current study employs Mueller-Hilton agar-based assays to evaluate several types of leaves of Vitex negundo Linn. (Nirgundi) for in vitro antibacterial activity against gram-positive (Staphylococcus aureus) and gram-negative (Escherichia coli) bacteria (disk diffusion method).The results suggested that the ethanolic extract of Vitex negundo showed zones of inhibition of 12.19 ± 0.445 mm, 17.23 ± 0.761 mm and 21.03± 0.559 mm against Staphylococcus aureus and 09.21± 0.357, 13.88 ± 0.652 and 18.48 ± 0.340 against E coli at concentrations of 100 g/mL, 200 g/mL and 400 g/mL, respectively. The percentage zone of inhibition of alcoholic extract of Vitex negundo 400 g/mL showed 74.93% and 68.95% against S. aureus and E coli, which is significant (P < 0.01) as compared to the positive control group. The aq. extract of Vitex negundo 400 g/mL inhibited S. aureus and E. coli with a significant (P< 0.01) zone of inhibition of 64.22 percent and 52.13 percent, respectively. The findings revealed that the ethanolic extracts of Vitex negundo Linn. Leaf showed a more prominent antibacterial effect than the aqueous extract against S. aureus and E. coli. Respectively.
