Abstract: PHBs are storage material synthesize by different microorganisms.The present study was focused on Production of PHB by bacterial isolates isolated from local soil samples.All the bacterial isolates were screened for their PHB producing ability and two isolates showing highest PHB production were selected for further studies.Optimization of different variables like pH, temperature, incubation period, carbon and nitrogen sources were performed.The maximum PHB accumulation was found when the medium was supplemented with glucose and sucrose as a carbon source. Yeast extract and peptone were found optimum nitrogen source.The optimum pH and temperature were 6.5-7.0 and 35°C respectively. The polymer can be further purified by dissolving in chloroform and precipitating with cold ethanol. PHB extraction carried out using sodium hypochlorite method exhibited a good PHB yield amounting to 73% and 80% for S1b and S3b isolates respectively. The extracted compounds on FTIR analysis gave characteristics functional units as C-H, CH2 ,-OH,C=O groups. UV-Vis spectrophotometric analysis provide confirmed similarity of extracted PHB and its suitability for bioplastic production.
